摘要:
本研究应用组织工程学技术构建了具有完整表皮和真皮结构的组织工程化大鼠皮肤.在电化学工作站中,按照吡咯单体浓度0.05 mol/L、聚合时间500 s、电流10 mA的条件制备聚吡咯膜.采用胰蛋白酶消化法从新生SD大鼠皮肤中分离获得角朊细胞并种植于聚吡咯膜上,导电培养获得较多数量的角朊细胞.将Ⅰ型胶原蛋白醋酸溶液与大鼠皮肤成纤维细胞相混合培养14 d,形成胶原蛋白-成纤维细胞凝胶真皮模型.将角朊细胞种植于真皮模型上浸没式培养5 d后,继续气-液面培养14 d.经固定、切片与染色,在光镜和扫描电镜下可见所构建的真皮模型与新生大鼠真皮具有类似的结构; 而大鼠皮肤模型则具有与正常新生大鼠皮肤相类似的结构,包括完整的真皮层和表皮层,表皮中含有完整的基底层、棘细胞层、颗粒层与角质层.该模型的建立,为组织工程化人皮肤的构建和临床应用奠定了基础.
Abstract:
A model of tissue engineered rat skin with dermal and epidermal structures is reconstructed. The conductive polypyrrole membranes were made in electrochemical station. The rat keratinocytes were isolated and planted on the membranes and large quantities of undifferentiated keratinocytes were then obtained after 72 h-culture of electrical stimulation. In the reconstruction of tissue engineered rat dermis, the rat dermal fibroblasts mixed with typeⅠcalf collagen acetic acid solutions were cultured 14 d and a dermal tissue-like structure of fibroblast-populated collagen gel matrix was formed. The reconstructed rat dermis showed a similar morphology to that of normal newborn rat dermis under microscopy. The keratinocytes were then seeded on reconstructed rat dermis. After a period of 5 d submerged culture, the cells were raised to air-liquid interface to culture for 14 d, a piece of reconstructed rat skin was formed. The reconstructed rat skin was similar in morphology to that of normal newborn rat skin with dermis and epidermis, which contained basal layer, granular layer and stratum corneum. The results are helpful to reconstruction and clinical application of tissue engineered human skin.
