摘要:
采用司盘80(Span 80)和吐温20(Tween 20)对茶多酚(TP)脂质体进行修饰,优化TP脂质体的制备条件,考察不同性质的表面活性剂对TP脂质体的制备及透皮性能的影响.试验表明:用大豆卵磷脂制备的TP脂质体优于蛋黄卵磷脂制备的TP脂质体; 以120 mg大豆卵磷脂、30 mg胆固醇、20 mg Span 80和20 mg Tween 20的配方制备的TP脂质体粒径最小(247.2 nm),且稳定性良好.增大Span 80的用量会使TP脂质体制备困难,同时使TP脂质体的粒径变大,但能显著提高TP脂质体的透皮吸收性能; 增大Tween 20用量使制备容易,同时可以显著减小脂质体的粒径,但当Tween 20用量超过20 mg或用量超过Span 80用量时,制备的TP脂质体不够稳定,且TP脂质体的透皮性能下降.
Abstract:
Span 80 and Tween 20 were investigated to explore their influence on particle sizes and percutaneous absorption of tea polyphenols(TP)liposomes. TP liposomes were prepared using soybean phosphatidylcholine and cholesterol. The particle size distribution was measured using photon correlation spectroscopy. The mean particle size of TP liposome which remained stable over 3 months storage was 247.2 nm under optimal experimental conditions with 120 mg phosphatidylcholine, 30 mg cholesterol, 20 mg Span 80 and 20 mg Tween 20. It was found that increasing the amount of Span 80 led to the decrease in the opportunity of successfully preparing TP liposomes and the increase in particle sizes. On the contrary, increasing the amount of Tween 20 facilitated the preparation and evidently reduced the mean particle sizes. However, when the amount of Tween 20 was more than 20 mg or the amount of Span 80, the TP liposomes were unstable. The percutaneous absorption experiments of TP liposomes through the rabbit skin were conducted using self-made Franz diffusion cell in vitro. When more Span 80 was added, the percutaneous absorption ability of TP liposomes was enhanced; when more Tween 20 was added, the percutaneous absorption ability was decreased. It is concluded that the introducing surfactants such as Span 80 and Tween 20 have significant influence on the preparation and percutaneous absorption of TP liposomes.
