摘要:
采用MTT实验评价自制Mn0.5Zn0.5Fe2O4纳米粒浸提液体外细胞毒性; 溶血实验评价其有无溶血作用和LD50评价其动物急性毒性,微核实验评价其有无致畸、致突变作用等.兔肝脏穿刺注射Mn0.5Zn0.5Fe2O4纳米粒,观察其对兔肝肾等组织和功能的影响.MTT结果显示:该材料对L-929细胞毒性为0~1级; 溶血实验中Mn0.5Zn0.5Fe2O4复合纳米粒溶血率为0.104%,远小于5%; 昆明小鼠腹腔注射该材料混悬液,其LD50为6.026 g/kg,其95%的可信区间为4.78~7.60 g/kg.微核实验结果表明:该材料对小鼠骨髓微核形成率与阴性对照组相比无显著差异,而与阳性对照组CTX组相比有明显差异.兔肝脏穿刺注射Mn0.5Zn0.5Fe2O4纳米粒心,肝、脾、肺、肾组织形态和肝、肾等血生化指标均无明显改变.结果表明自制的Mn0.5Zn0.5Fe2O4纳米粒具有良好的生物相容性.
Abstract:
MTT assay was used to evaluate the in vitro cytoxicity; Haemolysis test was carried out to estimate whether the ferrite had blood toxicity; LD50 was used to evaluate the in vivo acute toxicity; Micronucleus test was reckoned to identify whether it had short-time genetic toxicity; And morphology and blood biochemical test was used to evaluate the effect of Mn0.5Zn0.5Fe2O4nanoparticles on morphology of main organ tissues and the function of liver and kidney after Mn0.5Zn0.5Fe2O4nanoparticles were injected into rabbit liver tissue. The result of MTT assay shows that the toxicity of materials on mouse fibroblast(L-929)cell lines is between 0 to 1 degree approaching to no cytoxicity; In the haemolysis test, the haemolysis rate is 0.104% which is far less than 5%; LD50 is 6.026 g/kg and the 95% confidence interval is 4.78 to 7.60 g/kg. In the micronucleus test, the result shows no significant difference(P>0.5)compared with negative control group, while had significant difference(P<0.5)compared with positive control group; and the morphology and blood biochemical test shows that Mn0.5Zn0.5Fe2O4 nanoparticles have no obvious negative effect on morphology of main organ tissues, all blood biochemical indicators of experimental group have no significant difference(P>0.05)compared with negative control group and self-group before treatment. The results of our experiment indicate that Mn0.5Zn0.5Fe2O4 nanoparticles are a kind of materials with good biocompatibility.
