摘要:
为了得到超声辅助提取补骨脂素工艺的最佳工艺条件及数学模型,采用单因素实验和响应面法,对影响超声辅助提取补骨脂中补骨脂素工艺的因素进行分析.得到最佳的超声提取工艺:补骨脂的颗粒为100~120目,超声波的频率为40kHz,超声提取时间为41min,溶剂为体积分数80%的乙醇水溶液,料液比为10mL/g,溶剂的初始温度为52℃. 最佳工艺条件下补骨脂素的提取率可达0.621%,提取率远高于普通溶剂提取率.用Design-Expert 7.0.0软件对实验数据进行二次拟合及分析,建立了可靠、稳定的数学模型,并得到各因素对提取率的影响顺序由强到弱依次为溶剂起始温度、料液比、超声时间.
Abstract:
In order to obtain the optimal parameters of ultrasonic extraction of psoralen and establish corresponding mathematical model, single factor experiments and the response surface method are applied to measure the influence factors in the process of psoralen extraction from Psoralea pubescence with ultrasonic assistance. The optimal extraction conditions are obtained as follows: the mesh of grain is 100 to 120, the ultrasonic frequency is 40 kHz, the extraction time is 41 min, the solvent is 80% volume fraction of ethanol, the ratio of liquid to solid is 10 mL/g and the temperature is 52℃. Under these circumstances the optimized psoralen extraction yield can reach 0.621%, which is much higher than that obtained by conventional solvent extraction. A reliable and stable mathematical model is obtained by quadratic fitting and analysis of experimental results with Design-Expert 7.0.0. With this model, it is concluded that the influence of initial temperature, ratio of liquid to solid, and ultrasonic time on extraction yield decreases in order.
